Two-Site Immunochemiluminometric Assay for Human a1-Fetoprotein

چکیده
In this two-site immunochemiluminometric assay for human a1-fetoprotein, acndinium ester-labeled monoclonal antibodies are used that have an average incorporation ratio of 0.3 mol of acridinium ester per mole of antibody. The solid-phase antibody consists of sheep anti-a1-fetoprotein IgG covalently coupled to a diazonium derivative of reprecipitated aminoaryl cellulose. The assay involves a 1-h incubation after the simultaneousincubationof labeled and solid-phaseantibodies. The subsequentquantificationtime is 10 s per tube. The assay has a working range of 20 to 246 kilo-mt. units/L for CVs 10% and a sensitivity of detection of 1.3 kilo-mt. units! L. Experiments with monoclonal antibodies labeled to an average incorporation ratio of 2.8 mol of acridinium ester per mole of antibody suggest that assay sensitivity increases with increased specific activity. a1-Fetoprotein concentrations in the sera of pregnant women at 14-20 weeks of gestation, as measured by the present assay, agreed with the resultsof a conventionalradioimmunoassay.

Two-site immunochemiluminometric assay of intact human parathyrin in serum with use of a tracer peptide purified by reversed-phase high-performance liquid chromatography.
For this two-site immunochemiluminometric assay of intact human parathyrin (hPTH), the luminescent tracer was synthetic hPTH(53-84), conjugated via succinimide linkage to (aminobutyl)ethyl-isoluminol hemisuccinimide (abei-h). Purification of the labeled hPTH(53-84) by reversed-phase high-performance liquid chromatography allowed isolation of the conjugate having the highest incorporation of abe...
متن کاملMeasurement of choriogonadotropin by chemiluminescence immunoassay and immunochemiluminometric assay: 1. Use of isoluminol derivatives.
We have developed immunoassays, monitored by the detection of chemiluminescence, for measuring choriogonadotropin in human urine. These methods involve the use of derivatives of isoluminol and include: (a) a labeled antigen with a second antibody covalently linked to polyacrylamide beads as the solid-phase reagent (i.e., solid-phase chemiluminescence immunoassay); and (b) an excess concentratio...
متن کاملProinsulin level in diabetes mellitus measured by a new immunochemiluminometric assay.
Proinsulin, the precursor of insulin and C-peptide, is detectable in the circulation and is a potential marker of beta-cell dysfunction. Currently, circulating proinsulin can be measured accurately without the interference of insulin or C-peptide by immunometric assays and a few specific radioimmunoassays. An immunochemiluminometric assay was developed in our laboratory by using two immunopurif...
متن کاملIn vivo Characterization of Fusion Protein Comprising of A1 Subunit of Shiga Toxin and Human GM-CSF: Assessment of Its Immunogenicity and Toxicity
Background: Most cancer cells become resistant to anti-cancer agents. In the last few years, a new approach for targeted therapy of human cancer has been developed using immunotoxins which comprise both the cell targeting and the cell killing moieties. Methods: In the present study, the recombinant Shiga toxin A1 subunit fused to human granulocyte-macrophage colony stimulating factor (A1-GM-CSF...
متن کاملMultisite immunochemiluminometric assay for simultaneously measuring whole-molecule and amino-terminal fragments of human parathyrin.
The immunochemiluminometric assay described uses immobilized anti-human parathyrin (parathyroid hormone, hPTH)(1-44) and anti-hPTH(44-68) antisera and acridinium ester-labeled anti-hPTH(1-34) to simultaneously measure both intact hPTH and its amino-terminal fragments. Results by the assay correlate well with those by a cAMP-based bioassay and the Nichols Allegro immunoradiometric assay. The min...
